Toxicity assessment of tetrachloroethylene, trichloroethylene and 1,1,1-trichloroethane using human and monkey cells
The toxicity of tetrachloroethylene (1271847), trichloroethylene (79016), and 1,1,1-trichloroethane (71556) (TCE) to human and monkey cells was studied in-vitro. Cultures of human KB cells and African-Green-monkey kidney (AGMK) cells were exposed to tetrachloroethylene, trichloroethylene, or TCE for 72 hours. Doses ranged from about 100 to 700 micrograms per milliliter (microg/ml). Inhibition of growth was used as the index of toxicity. Dose effect curves were constructed. Both KB and AGMK cells showed dose/dependent inhibition of growth when treated with the compounds. There was no clear distinction in sensitivity between KB and AGMK cells to tetrachloroethylene, or TCE, and trichloroethylene. The doses for 50 percent inhibition of growth (ID50) in the KB cultures were tetrachloroethylene 195microg/ml, TCE 420microg/ml, and trichloroethylene 630microg/ml. The ID50s in the AGMK cells were PERC 140microg/ml, TCE 380microg/ml, and trichloroethylene 550microg/ml. The authors conclude that tetrachloroethylene seems to be more more toxic than trichloroethylene and TCE to KB and AGMK cells. The results also show that TCE is about five times more toxic than trichloromethane, studied previously in KB and AGMK cells.
