Cytochrome P450 isoform distribution in livers of Wistar-rats treated with phenobarbital (50066) (PB), 3-methylcholanthrene (56495) (MC), Aroclor-1254 (11097691) (AR), pregnenalone-carbonitrile (PC), imidazole (288324) (IM), and beta-naphthoflavone (6051872) (NF) was studied. Monoclonal antibodies developed in BALB/c-mice immunized with cytochrome P450 from rats pretreated with PB, MC, or AR were used for immunoblots of electrophoretically separated microsomes or for immunohistochemical staining of sections of treated rat liver. Initial studies with these antibodies suggested that PB induced the isoforms P450b, P450PB, and P450 alpha. MC appeared to induce the isoforms P450c and P450d. Monoclonal antibodies were tested against microsomes and liver sections from rats treated with 80mg/kg PB, 20mg/kg MC, 500mg/kg AR, 80mg/kg PC for 3 days, 200mg/kg IM for 3 days, and 80mg/kg NF for 3 days. Microsomes from rats treated with PC reacted with antibodies to P450 isoforms induced by PB, while those from NF treated rats reacted with MC induced isoforms. IM isoforms appeared to be unique in this group of compounds, while AR treated liver microsomes reacted with all monoclonal antibodies. Immunohistochemical analysis demonstrated a gradient of staining, with more intense staining in cells nearer to the central veins than the portal veins.
