Abstract
Cholinesterase antagonist monitor (CAM) methods were used to detect high molecular weight chlorinated hydrocarbons. Cholinesterase was replaced in the method design by lactate-dehydrogenase (LDH). About 70 inhibition assays were performed on seven representative test compounds: aldrin (309002), toxaphene (8001352), DDT (50293), aroclor-1242 (53469219), pentachlorophenol (87865), 2,4,5-trichlorophenoxy-acetic-acid (93765), and lindane (58899). Spectrophotometric determination of the absorbance of the reaction mixture at the 340 nanometer peak of NADH was used to determine the rate of the reaction in these experimental studies. Concentrations of at least five chlorinated hydrocarbons were possible at levels of 0.0004 to 0.000006 molar in a 20 percent methanol/water solution. Interference by nonchlorinated analogs is not expected to occur. The method has potential use in field situations. Methods to immobilize the enzyme on a suitable support are needed to make reuse of the enzyme possible and simplify the detection procedure. It is also important to design an extraction, concentration procedure which will precede the use of the LDH detection method when trying to identify chlorinated hydrocarbons with low water solubility.
